چکیده

Abstract Background Cigarette smoke (CS) exposure increases corticosteroid insensitive asthma related to increased Th17 phenotype, and new treatment strategies are needed for CS-associated asthma. Histone deacetylase 2 (HDAC2), found in the airway epithelium, is critical ameliorating glucocorticoids insensitivity. We recently demonstrated anti-inflammatory effects of CpG oligodeoxynucleotides (CpG-ODNs) on CS-exposure However, CpG-ODNs HDAC2 expression enzymatic activity remain unclear. This study aimed assess whether protect against excessive immune responses CS-induced through HDAC2-dependent mechanisms compared their with those corticosteroids. Methods The alone combination budesonide (BUD) inflammation Th2/Th17-related were determined using an vivo model cultured bronchial epithelial (HBE) cells administered ovalbumin (OVA) and/or cigarette extract (CSE). retinoid-related orphan nuclear receptor γt (RORγt) also assessed mouse lung specimens HBE cells. Results BUD synergistically attenuated CS asthmatic by modulating influx eosinophils neutrophils, remodeling, Th2/Th17 associated cytokine chemokine production, hyperresponsiveness blocking RORγt-mediated induced expression/activity. In vitro , synergized inhibit production OVA- CSE-challenged while suppressing RORγt increasing Conclusions reversed downregulation enhanced sensitivity CS-exposed mice CSE-induced glucocorticoid treatment. effect may be restoration via RORγt/IL-17 pathway regulation, suggesting that potential corticosteroid-sparing agents use Th17-biased conditions.

Histone deacetylase 2 is phosphorylated, ubiquitinated, and degraded by cigarette smoke.

Cigarette smoke (CS)-induced lung inflammation involves the reduction of histone deacetylase 2 (HDAC2) abundance, which is associated with steroid resistance in patients with chronic obstructive pulmonary disease and in individuals with severe asthma who smoke cigarettes. However, the molecular mechanism of CS-mediated reduction of HDAC2 is not clearly known. We hypothesized that HDAC2 is phosp...

Immunotherapy of Chenopodium Album Induced Asthma by Intranasal Administration of CpG Oligodeoxynucleotides in BALB/c Mice

Background: There are many therapeutic methods for allergic conditions. CpG oli-gonucleotides play a critical role in immunity via the augmentation of Th1 and suppres-sion of Th2 responses. Objective: In the present study we aimed to estimate the effec-tiveness of intranasal administration of CpG ODN plus Chenopodium album allergen in allergic asthma compared with the administration of allergen...

Correlation between cigarette smoke exposure and prevalence of asthma and allergy symptoms in children, Yazd

Introduction: Cigarette smoke is one of the most prevalent pollutants. Regarding the development of lungs and immune system at early stages of life, exposure to these substances can have adverse effects in this period. It seems that tobacco smoke is not an etiology for wheezing attacks burden, instead, it is an exacerbating factor. This survey was designed to determine the relation of environme...

Histone deacetylase (HDAC) inhibitors attenuate cardiac hypertrophy by suppressing autophagy.

Histone deacetylases (HDACs) regulate cardiac plasticity; however, their molecular targets are unknown. As autophagy contributes to pathological cardiac remodeling, we hypothesized that HDAC inhibitors target autophagy. The prototypical HDAC inhibitor (HDACi), trichostatin A (TSA), attenuated both load- and agonist-induced hypertrophic growth and abolished the associated activation of autophagy...

CpG oligodeoxynucleotides enhance FccRI-mediated cross presentation by dendritic cells

Dendritic cells (DC) can trigger naive CD8 T cell responses by their capacity to cross-present exogenous antigens via the major histocompatibility complex class I pathway. The myeloid class I IgG receptor, FccRI (CD64), is expressed on DC, and in vivo targeting of antigens to FccRI induces strong humoral and cellular immune responses. We studied the capacity of human FccRI (hFccRI) to facilitat...
